amount ofは数えられない名詞の「量」、number ofは数えられる名詞の「数」を表します。 amount of water（ある量の水）のように量として測るものにはamount、the number of students（生徒の数）のように一人、二人と数えるものにはnumberを使います。
amountは水や時間などを一つのまとまりとして測る「量」、numberは人や本などを一つずつ数える「数」です。まず後ろの名詞を数えられるかどうかで判断しましょう。

基本は可算名詞と不可算名詞の違いと同じです。water、information、work、trafficなどは通常は数えないためamount、books、people、mistakes、ordersなどは数えられるためnumberを選びます。
amount ofの意味と使い方｜数えられない名詞の量
amountは「総量」「分量」という意味です。後ろには、直接1つ、2つと数えない不可算名詞を置きます。
- We need a small amount of water.
少量の水が必要です。 - The amount of information is increasing.
情報量が増えています。 - She spent a large amount of time on the project.
彼女はその企画に多くの時間を費やしました。 - Reduce the amount of sugar you eat.

a large amount ofは「大量の」、a small amount ofは「少量の」です。会話ではa lot ofも広く使われますが、amount ofは量そのものを意識するやや説明的・事務的な表現です。
number ofの意味と使い方｜数えられる名詞の数
numberは「数」「個数」を表し、後ろには可算名詞の複数形を置きます。
- The number of visitors is increasing.
訪問者数が増えています。 - We received a large number of orders.
多数の注文を受けました。 - Only a small number of people agreed.
賛成した人は少数でした。 - Please enter the number of items.
品数を入力してください。
number ofの後ろを単数形にしない点に注意してください。× the number of student ではなく、○ the number of studentsです。

the amount ofとthe number ofの動詞は単数形
the amount of＋名詞とthe number of＋複数名詞が主語になる場合、中心はamountまたはnumberという一つの量・数値なので、通常は単数動詞を使います。
- The amount of waste is decreasing.
廃棄物の量は減っています。 - The number of errors has fallen.
エラー数は減りました。
一方、a number of＋複数名詞は「多数の〜」という別の決まりで、複数動詞を使います。この違いはa number ofとthe number ofの違いで詳しく整理しています。
迷ったら「何個あるか」を答えるならnumber、「どのくらいあるか」を答えるならamountと考えてください。moneyは数えない名詞ですが、dollarsやyenという単位にすればnumberで表せます。
